No laboratório liderado por Jonathan Howard estudam os mecanismos de resistência ao parasita protozoário intracelular, Toxoplasma gondii, um parente da malária, que infeta cerca de 40% da espécie humana. O grupo de investigação foca-se no estudo da imunidade de ratinhos contra T. gondii porque os hospedeiros primários do parasita, no qual produz gametas e faz meiose, são gatos. Portanto, o ciclo de vida de T. gondii e a sua abundância no meio ambiente, são causados por um ciclo infeccioso entre gato e rato.
A imunidade dos ratinhos contra T. gondii assenta num mecanismo ausente em humanos, a ação das GTPases induzidas por interferão gama (proteínas IRG), que , de forma cooperativa, destroem o vacúolo no qual o parasita vive. Este mecanismo, por sua vez, é alvo de parasita virulentos, que dispõem de uma família de quinases responsáveis pela inativação das proteínas IRG.Tanto as proteínas IRG como as quinases são maciçamente polimórficas, consistentes com uma dinâmica co-evolutiva complexa.
O trabalho deste grupo de investigação abrange estudos ecológicos em ratinhos selvagens, estudos biológicos, bioquímicos e estruturais.
