Transgenics: Mouse

The Mouse Transgenics Unit is a core facility that provides transgenic and gene targeted mice to investigators of IGC, as well as to investigators within Portugal and abroad. For externals is fee-for-service based. We are competitive in success rates and pricing.
The Unit performs microinjections of DNA into fertilized embryos to generate transgenic mice, microinjections of gene-targeted ES cells into host blastocysts to generate gene knock-out or knock-in mice and of CRISPR/Cas9 and gRNAs as well as DNA donors into mouse zygotes for gene editing projects, using the ultimate technologies in the field. We provide users general consultations.

The researchers provide the DNA constructs, gRNAs or ES cells. The TU staff then performs all microinjections and surgical procedures required for the production of the mutant mice. We deliver founders or chimeras that are then further crossed and analyzed by the requesting researchers.

SERVICES

The Transgenics Unit is dedicated to producing genetically modified mice via transgenic and gene-targeting methodologies using state-of-the-art techniques.

Production of transgenic mice by pronuclear DNA microinjection using both conventional expression constructs and Bacterial Artificial Chromosomes (BACs) in FVB/N and C57Bl/6 genetic backgrounds.

Introduction of targeted modifications into endogenous genomic loci following conventional embryonic stem (ES) cell-mediated approaches and with the CRISPR/Cas9 technology in FVB/N and C57Bl/6 genetic backgrounds.

Transgenic and gene-targeted mice in Germ free/Gnotobiotic conditions, both in C57Bl/6 and FVB/N backgrounds, in collaboration with the Axenic/Gnotobiology Facility
