Branching morphogenesis of neuronal dendrites: Mechanisms, models and microtubules

Branched networks distribute information, nutrients, and forces through cells, tissues, and organisms. In the nervous system, the highly ramified arbors of dendrites provide the substrate for the high connectivity of the brain. To elucidate the principles underlying branching morphogenesis in neurons, we have visualized the growth of dendrites throughout larval development of Drosophila sensory neurons with high temporal and spatial resolution. We discovered that dendrite tips undergo dynamic instability, transitioning rapidly and randomly between three states - growing, shrinking, and paused. A computational model based on the measured tip dynamics predicts the coarse and fine structures of the dendrites, including their variations, showing that the complex and high variable morphologies of these cells are a consequence of the stochastic dynamics of their tips. We are currently exploring the roles of microtubules, motors and microtubule regulators on dendrite morphogenesis.
